Reverse arrays
typically use multi-samples of extracts from cells or tissues or
biofluids such as blood, serum, cerebral spinal fluid, milk, nipple
aspirates, urine, semen, and tears, for probing with a primary
antibody. This is ideal for situations where a promising lead
antibody from an antibody microarray analysis has been validated by
Western blotting and shown to be monospecific in the biospecimen
type of interest.
At this stage, it is
now necessary to confirm the reproducibility of the identified
biomarker in a large number of samples, for example, in diverse
patients with a disease and healthy controls. With replicate copies
of the arrayed samples, it is feasible to efficiently screen the
same samples with a panel of lead antibodies, one antibody tested on
each array copy.
The availability of
our Custom Lysate Array services completes our unique biomarker
discovery platform from initial identification of promising
biomarkers and antibodies to detect them, through to immunoblotting
validation as confirmed target proteins, to finally biological
validation as statistically significant distinguishing biomarkers
across a large cohort of samples.
